Color Movies with VoxBlast: 3D Reconstruction of Point Spread Function from 0.2 Micron Bead

This movie shows a 3D reconstruction of a point spread function (PSF) from a microscope. To create the dataset, a .2 micron fluorescent bead was imaged under an Olympus BH2 microscope. About 40 images were taken with a Photometrics cooled CCD camera and 100x 1.4 N.A. lens. The images were taken about .2 microns apart.

The stack of images were then put into VoxBlast. A color palette and opacity function was created to produce a semi-transparent view of the PSF. Note the slight astigmatism in the lens in the top view.

The images that were captured could be used with MicroTome and the constrained iterative algorithm to deconvolve a stack of specimen images captured under similar circumstances.
