Fluorescence in Situ Hybridization (FISH) with DNA: Greater Accuracy for Automated Spot Counting

After Haze Removal

DNA probes locate precise locations on chromosomes and adhere to them with an antibody. The probes fluoresce under a microscope and indicate the presence of certain characteristics in the DNA. For example, a researcher can determine the gender of a child, the presence or absence of genetic defects, or the presence of pathological conditions affecting DNA, such as cancer.

Manually locating and counting the probes is time consuming. Computer programs that can count and measure spots are often used to accelerate the process. The accuracy and speed of such programs is determined, in part, by the clarity of the images. The computer must be able to distinguish the probes from the background and separate overlapping probes from one another.

As illustrated in the image to the left, the contrast between background and probe is significantly improved with digital haze removal techniques. In addition, there is an increase in resolution that helps to separate overlapping probes.

VayTek's proprietary Single Image Haze Removal algorithm is particularly suited for this application. Pinhole based scanning confocal microscopes block too much light and lack the sensitivity to capture sufficient information.
