One of the advantages of digital haze removal is that images
can be captured using any appropriate light source. There are
several implications of this point:
- It is not necessary to use lasers. Conventional light sources
can be used. This can result in less photobleaching.
- Wavelengths not available with lasers can be used. For example,
ultra violet sensitive dyes, such as fura, are appropriate.
- Using filters it is possible to tune excitation wavelengths
precisely to the dyes being used.
- A laser source can be used, if desired. A pulsed laser can
be used in some instances which practically eliminates photobleaching.
The example image shows a leech embryo cell that was micro-injected
with a rhodamine dextran conjugate dye which photobleached quickly.
The parent cell, in the upper right, divided and produced the
daughter cell in the lower left. The daughter cell had a smaller
amount of the dye and was even more difficult to image.
When a laser scanning confocal was used, the specimen bleached
after one scan. Using a conventional light source, a shutter
and the proper filters, eleven images at different vertical depths
were acquired before the dye quenched. The digitally processed
image revealed more detail than the single laser scanned image.
